A UV-Vis detector works by passing the sample eluent from the HPLC column through a flow cell that is illuminated by a beam of UV or visible light. The sample eluent absorbs some of the light, and the amount of absorption is measured by a detector. The detector converts the absorption signal into an electrical signal, which is then processed by a computer. The computer generates a chromatogram, which is a plot of the detector signal versus time. The chromatogram shows the peaks that correspond to the different components of the sample.
